Inhibition of inflammatory factors by parthenolide in human renal mesangial cells under hyperglycemic condition

摘要: The aim of this study was to investigate the anti-inflammatory effects parthenolide (PTN) in human renal mesangial cells (HRMCs) under high ambient glucose conditions. First we determined noncytotoxic concentration PTN HRMCs by performing MTS assay. Enzyme-linked immunosorbent (ELISA) analysis performed determine expressions interleukin (IL)-1, IL-18, tumornecrosis factor (TNF)-, transforming growth (TGF)-1, monocyte chemoattractant protein (MCP)- 1, macrophage inflammatory (MIP)-1, RANTES and prostaglandin (PG)E2. total nitric oxide (NO) Griess reaction. Treatment with less than 50 mol/L did not affect viability HRMCs, while more 100 imol/L concentrations markedly altered cell viability. In present study, treatment mmol/L increased level IL-1, TNF-, TGF-1, MCP-1, MIP-1, RANTES, PGE2 NO. remarkably abolished increase these molecules a dose-dependent manner. Moreover, 20 almost completely reversed IL-1 TNF- expression, expression RANTES. conclusion, can inhibit high-glucose-induced cytokines HRMCs. Hence, may be considered promising drug potent effect addition its strong anticancer, anti-angiogenesis, antineurodegenerative effects.
