摘要: Lymphocyte-derived cytokines induce sequential expression of monocyte- and T cell-specific chemokines in human mesangial cells. Chemokines are a family small related proteins that play an important role the selective recruitment different leukocyte populations to sites inflammation. Human glomerular cells potent producers variety chemokines. Here we examined kinetics cell chemokine with focus on C-C or β monocyte chemoattractant protein-1 (MCP-1), regulated upon activation, normal expressed secreted (RANTES), macrophage inflammatory protein-lα (MIP-1α), C-X-C α interleukin-8 (IL-8) response lymphocyte- monocyte-derived growth factors. It was found interferon-γ (IFN-γ), cytokine produced by TH1 lymphocytes, synergized tumor necrosis factor-α (TNF-α) RANTES IL-1β MCP-1 synthesis. Time course studies revealed early peak mRNA monocyte-specific activation TNF-α contrast RANTES, which reached highest level after 18 hours. This sequence TNF-α-induced confirmed protein level. As another T-lymphocyte specific chemokine, MIP-1α only plus IFN-γ similar those expression. Finally, unlike other factors basic fibroblast factor (bFGF) induced MCP-1, IL-8 expression, suggesting involvement autocrine regulation mechanisms
