摘要: There is a raising demand for sensitive and high throughput MS based methods screening purposes especially tailored to the detection of allergen contaminants in different food commodities. A challenging issue represented by complex matrices where antibody-based kits commercially available might encounter objective limitations consequently epitope masking phenomena due multitude interfering compounds arising from matrix. The performance method duly optimized extraction simultaneous soy, egg milk allergens cookie matrix microHPLC-ESI-MS/MS, herein reported. Thanks innovative configuration versatility shown dual cell linear ion trap used, most intense reliable peptide markers were first identified untargeted survey experiment, subsequently employed design an ad hoc multi-target SRM method, on transitions recorded each selected precursor peptide. sample purification protocol was also including additional step sonication, which resulted considerable improvement peptides. Data Dependent™ Acquisition scheme allowed fill out tentative list potential markers, further filtered upon fulfilling specific requirements. total eleven peptides monitored simultaneously confirmation allergenic contaminant two markers/protein order retrieve quantitative information. Relevant LODs found range 0.1μg/g 0.3μg/g 2μg/g soy.
