摘要: MutS, a DNA mismatch-binding protein, seems to be promising tool for mutation detection. We present three MutS based approaches the detection of point mutations: retardation, protection mismatched against exonuclease digestion, and chimeric proteins. retardation in polyacrylamide gels stained with SYBR-Gold allows using 1–3 µg Thermus thermophilus his 6 -MutS protein 50–200 ng PCR product. The method enables search broad range from single up several nucleotide, as mutations over nucleotides could detected electrophoresis without due mobility
