摘要: Suspensions of human embryonic stem cell-derived retinal pigment epithelium (hES-RPE) and fetal RPE (fRPE) resurface aged age-related macular degeneration (AMD) Bruch’s membrane (BM) to a limited degree at day 21 in organ culture. The presence AMD-related changes, such as basal linear deposit, is associated with worse BM resurfacing. Survival differentiation hES-RPE fRPE on or AMD enhanced greatly (200 %) if biologically synthesized extracellular matrix (ECM) (bovine corneal endothelial cell (BCEC) ECM) laid down prior transplantation. Transplanted survival even more (400–1,000 treated BCEC conditioned medium during culture BM. Cells so also produce variety neurotrophic factors (e.g., brain-derived factor, epithelium-derived nerve growth factor) while they do not exhibit highly differentiated morphology. This factor production may contribute improved vision that has been observed some patients who have received allogeneic transplants. Future efforts are focused identifying the bioactive components BCEC-CM this material can be reconstituted for clinical use an adjunct improve transplant eyes.
