摘要: A relatively simple method for immobilizing synthetic peptides as a receptor onto gold surface using the self-assembling monolayer (SAM) technique has been investigated. peptide with an amino acid sequence similar to 9-21 gD of herpes simplex virus type 1 was modified alkylthiol chain and adsorbed in combination alkyithiol without according SAM procedures. The resulting self-assembled layers (SARs) were able specifically interact monoclonal antibody directed against pep-tide. Binding studies monitored plasmon resonance setup showed that response depended on composition SAR; maximum reached at approximately 3 mol% coverage. itself exceeded value obtained from passive adsorption antibodies under conditions, indicating formed layer is highly oriented. equilibrium binding properties immobilized molecules found be identical those when interacted molecule. presence hydrophobic moiety derivates contributes ordering monolayer: less specific interaction occurred SARs used.
