摘要: A new E. coli expression system, the pHCE-IIB vector, using a constitutively expressing HCE promoter derived upstream from d-amino acid aminotransferase (d-AAT) gene of Geobacillus toebii, was developed for high-level foreign proteins without induction. The efficiency recombinant human tumor necrosis factor-α (rhTNFα) compared with that pET14B vector under control T7 induced by IPTG. total amount rhTNFα produced pHCE system approximately twice pET optimal fermentation conditions thereby demonstrating convenience and economical advantage constitutive over conventional vector.
