Turbidometric characterization of the postantifungal effect: comparative studies with amphotericin B, 5-fluorocytosine and miconazole on Candida albicans.

The phenomenon of persistent suppression Candida albicans yeast cell growth after short drug exposures (postantifungal effect; PAFE) was determined by performing comparative studies using different concentrations 5-fluorocytosine (5-FC), amphotericin B (AMPH) and miconazole (MCZ). An in vitro turbidometric method used to measure quantitate the PAFE removal dilution following exposure C. cells for 0.5 h, 1 h or 2 h. difference time (h) required control test cultures increase absorbance level antifungal agent. Zusammenfassung. der vorliegenden Studie wird die Unterdruckung des Hefezellwachstums von nach Antimyzetika-Kurzexposi-tion untersucht, sog. postantimyzetische Effekt (PAFE). Hierzu wurde 5-Fluorcytosin Am-photericin und Miconazol (MCZ) unterschiedlichen Konzentrationen eingesetzt. Zur Messung zur Quantifi-zierung Eliminierung Antimy-zetikums durch Herausverdunnen 0,5 oder Exposition albicans-Hefezellen eine turbidimetrische Methode be-nutzt. Der bestimmt uber Zeitdif-ferenz (h), welche Kontrollkultur bzw. Test-kulturen benotigten, um das Absorptionsniveau Antimyzetikums zu er-reichen. Bei alien gepuften Antimyzetika ein gefunden. Die Dauer hing sowohl Antimyzetika-Konzentration wie auch Expositionszeit ab. Eine fuhrt mit 0,6–16,7, 0–16,5 MCZ 0,1–14,1 den meisten Fallen fuhrte 1–2-h-Exposition Zellen bei einer leicht verlangerten PAFE. Langere konnten geringeren im Vergleich induziert werden. Ergebnisse solcher PAFE-Studien konnen fur Ausle-gung Behandlungsschemata nutzlich sein, da langere intermittierende An-wendung statt kontinuierlicher Antimyzetika-Gaben gestatten konnten.
