摘要: Tissue engineering approaches have been clinically tried to repair damaged articular cartilages. It is an essential step uniformly seed chondrocytes into 3D scaffolds in order reconstruct tissue-engineered cartilages vitro, but the tissue could not provided with efficient cell seeding methods. Type I collagen used and known as a cytocompatible material, having recognition sites for integrins. Collagen gel encapsulating has making regenerated cartilages, it found difficult keep its original shape after long-term culture, because of shrinking. On other hand, scaffolds, either nonwoven structure or sponge-like structure, involve difficulty that be seeded, although they adequate initial mechanical properties. In this study, by combining gelation PLLA scaffold, we achieved uniform scaffold. Bovine were mixed type solution, solution was poured scaffold (1.5 mm thick, diameter 15 mm). The collagen-chondrocyte mixture made at 37 degrees C 1 h. 0.39% viscous enough prevent cells from precipitating during gelation. Almost all able incorporated mixing subsequently gel, while 30-40% seeded suspension trapped scaffolds. method presented, where then serve alternative vitro cartilage regeneration, also simultaneously advantages both materials.
