摘要: Mesenchymal stromal/stem cells (MSCs) are widely distributed in different tissues such as bone marrow, adipose tissues, peripheral blood, umbilical cord and amnionic fluid. Recently, MSC-like were also found to exist rat olfactory bulb capable of inducing differentiation into mesenchymal lineages – osteocytes, chondrocytes adipocytes. However, whether these can differentiate myocardial is not known. In this study, we examined bulb-derived MSCs could vitro. Fibroblast-like isolated from the neonatal rats grown under four conditions: no treatment; presence growth factors (neuregulin-1, bFGF forskolin); co-cultured with cardiomyocytes; cardiomyocytes plus neuregulin-1, forskolin. Cell was monitored by RT–PCR, light microscopy immunofluorescence, western blot analysis contractile response pharmacological treatments. The fibroblast-like expressed CD29, CD44, CD90, CD105, CD166 but CD34 CD45, consistent characteristics MSCs. Long cylindical that spontaneously contracted only observed following 7 days co-culture RT–PCR indicated cylindrical markers, Nkx2.5, GATA4, sarcomeric α-actinin, cardiac troponin I, myosin heavy chain, atrial natriuretic peptide connexin 43. They contained sarcomeres gap junction sensitive treatments (adrenal cholinergic agonists antagonists). These findings indicate MSC myocardial-like cells. Copyright © 2013 John Wiley & Sons, Ltd.
