摘要: Publisher Summary This chapter focuses on the quantifying thrombin-catalyzed release of fibrinopeptides from fibrinogen using high-performance liquid chromatography (HPLC). It presents several methods in which HPLC analysis can yield (1) initial rates fibrinogen, (2) Miehaelis–Menten parameters catalytic constant( k CAT ) and (Michaelis constant) K M for a-thrombin-catalyzed fibrinopeptide A (FPA) fibilnopeptide B (FPB) fibrin I, (3) specificity constant / both FPA FPB at concentrations below , (4) inhibition hirugen, an exosite inhibitor α-thrombin. The also discusses profile coeluting activation peptides factor XIII. describes a method determining concentration α-thrombin that is catalytically competent to process fibrinogen.
