MicroRNA expression profiles associated with pancreatic adenocarcinoma and ampullary adenocarcinoma.

摘要: MicroRNAs have potential as diagnostic cancer biomarkers. The aim of this study was (1) to define microRNA expression patterns in formalin-fixed parafin-embedded tissue from pancreatic ductal adenocarcinoma, ampullary normal pancreas and chronic pancreatitis without using micro-dissection (2) discover new microRNAs combinations tissue. 664 170 adenocarcinomas 107 were analyzed a commercial assay. Results compared with pancreatitis, duodenal adenocarcinoma. In all, 43 had higher 41 reduced pancreas. 32 differently expressed adenocarcinoma (17 higher; 15 reduced). Several these not before been related diagnosis (eg, miR-492, miR-614, miR-622). MiR-614, miR-622, miR-135b miR-196 most expressed. MicroRNA profiles correlated (0.990). for described the literature consistent our findings, profile (miR-196b-miR-217) validated. We identified more significant profile, difference between miR-411 miR-198 (P=2.06 × 10(-54)) LASSO classifier 19 (sensitivity 98.5%; positive predictive value 97.8%; accuracy 97.0%). also subclassify into pancreatobiliary or intestinal type. conclusion, we found that two could roughly separate neoplastic non-neoplastic samples. A constructed which discriminate high sensitivity accuracy. Ongoing prospective studies will evaluate if are useful on fine-needle biopsies early cancer.
