摘要: Whether osteoarthritis (OA) is associated with alterations in the epigenetic status of anabolic factors largely unknown. To answer question, we investigated DNA methylation and histone modification SOX-9 gene promoter, a typical gene, articular cartilage from nine patients femoral neck fractures without OA hip patients. Methylation-specific PCR (MSP) bisulfite sequencing analysis (BSQ) showed that promoter was increased compared to normal cartilage. The decreased protein expression chondrocytes reversed by treatment 5-azacytidine (5-AzaC), demethylating agent. Methylation proximal promoters reduced binding affinity transcription CCAAT-binding factor/nuclear factor-Y cyclic adenosine monophosphate (cAMP) response element–binding. There significant increase H3K9 H3K27 trimethylation decrease acetylation H3K9, 15, 18, 23, 27 at chondrocytes. These findings suggest change including altered modification. © 2013 American Society for Bone Mineral Research.
