Expression of c-erbB receptors and ligands in human bronchial mucosa.

摘要: The epidermal growth factor receptor (EGFR, c-erbB1) plays a pivotal role in maintenance and repair of epithelial tissues; however, little is known about coexpression c-erbB receptors their ligands human bronchial epithelium. We therefore analyzed the expression these molecules cultured cells normal mucosa, using reverse transcription-polymerase chain reaction (RT- PCR), flow cytometry, immunohistochemistry. Messenger RNA (mRNA) encoding EGFR, c-erbB2, c-erbB3, but not c-erbB4, was detected primary cultures cells, as well epithelial-derived cell lines H292 16HBE 14o-. Transcripts (EGF), heparin binding (HB-EGF), transforming factor-alpha (TGF-alpha), amphiregulin (AR) were also detected, three four confirmed by immunocytochemical staining cells. Immunohistochemical analysis resin- or paraffin-embedded sections from surgical specimens mucosa revealed strong membrane for EGFR within epithelium; this particularly evident between basal aspect columnar patterns c-erbB2 c-erbB3 epithelium similar to those EGFR. Immunostaining EGF, TGF-alpha, AR, HB- betacellulin (BTC) intense submucosal glands; with exception BTC, ligand immunoreactivity observed epithelium, where it paralleled staining. Colocalization demonstrates potential productive interactions Further study may help define
