摘要: A modulating action of hyperforin (an active compound the extract from Hypericum perforatum) on a high-threshold component calcium current, sensitive to application 100 nM ω-Aga-IVA toxin and identified as P was studied freshly isolated Purkinje neurons with use patch-clamp technique in whole-cell configuration. It shown that extracellular 0.8 μM caused shift current-voltage (I-V) relationship current by -(8 ± 2) mV, slowdown activation kinetics, decrease amplitude this current. The I-V kinetics developed for less than 10 sec, while P-current decreased much longer time (several minutes) depended intracellular concentration Ca2+ ions. at completely blocked recorded inward presence hyperforin. In experiments perfusion neurons, we found interaction its binding site occurs external side cell membrane. study mechanisms involved hyperforin-induced modulation revealed 1 mM GTPγS (activating GTs proteins, well activating or blocking GMs proteins) 1-2 GDPβS (blocking solution did not affect Hyperforin-induced Ca2+-independent were abolished 0.5 calmidazolium medium.
