摘要: The hippocampus, a key structure in learning and memory processes, receives powerful cholinergic innervation from the septum contains nicotinic acetylcholine receptors (nAChRs). Early postnatal development, activation of nAChRs by nicotine or endogenous contributes to enhance synaptic signalling. Here, patch-clamp technique was used assess contribution alpha7 beta2-containing (alpha7* beta2*) nicotine-elicited modulation GABAergic glutamatergic activity at network single-cell level immature hippocampus wild-type (WT), alpha7-/- beta2-/- mice. We found that alpha7* beta2* were sufficient modulate nicotine-induced increase frequency spontaneously occurring giant depolarizing potentials (GDPs), which are generated synergistic action glutamate GABA, thought play crucial role neuronal wiring. However, but not essential interictal discharge recorded after day 3 presence bicuculline, when GABA shifted hyperpolarizing direction. To correlate these observations with changes transmission, we spontaneous postsynaptic currents pyramidal cells interneurons localized stratum oriens, pyramidale radiatum, slices obtained WT knock-out animals. early life exert fine regional transmission underlies synchronization.
