Liver plasma membrane: the source of high molecular weight alkaline phosphatase in human serum.

摘要: This study presents biochemical, histochemical, morphological and immunological evidence that part of the high molecular weight alkaline phosphatase observed in serum patients with liver disease particularly cases intrahepatic cholestasis or focal-, extrahepatic obstruction originates from plasma membrane. The protein complex contains several membrane enzymes behaves like a fragment after isopycnic density gradient ultracentrifugation sucrose, cesium chloride metrizamide. Electron microscopic examination revealed triple-layered vesicle which retained activity. Incubation human cells anti-serum against purified multienzyme resulted fixation antibodies on as shown by positive fluorescence. These fragments are not biliary origin.
