摘要: Summary Pluripotent stem cells can be differentiated into 3D retinal organoids, with major cell types self-patterning a polarized, laminated architecture. In static cultures, organoid development may hindered by limitations in diffusion of oxygen and nutrients. Herein, we report bioprocess using rotating-wall vessel (RWV) bioreactors to culture organoids derived from mouse pluripotent cells. Organoids RWV demonstrate enhanced proliferation, well-defined morphology improved differentiation neurons including ganglion S-cone photoreceptors. Furthermore, at day 25 (D25) reveal similar maturation transcriptome profile as those D32 culture, closely recapitulating spatiotemporal postnatal 6 retina in vivo . Interestingly, however, do not differentiate further under any in vitro condition tested here, suggesting additional requirements for functional maturation. Our studies that accelerate improve growth modeling disease evaluation therapies.
