摘要: Metagenomic and metatrascription technologies were developed rapidly applied widely in environmental microbiology recent years also gradually biohydrometallurgy. However, how to get good quality DNA RNA simultaneously is still the key factor determine success relibility of whole data analysis. In this study, a wide used extraction method [ was optimized for crude nucleic acid from acidic habitat. Our results showed that buffer (pH 7.0) contain PIPES salt could effectively extract (including RNA) four pure culture several samples. Qiagen isolation system (All prep DNA/RNA Mini kit) separate RNA. Good with A260/280 around 1.87±0.01 total 33.88±7.85 μg A260/230 2.07±0.07 20.00±2.73 obtained 300 mL A.f medium (strain concentration 2×107). All isolated be downstream molecular This study provides useful simultaneous extraction, separation purification habitat, which accelerate quantitative analysis biohydrometually therefore increase efficiency biohydrometually.
