摘要: The intact human growth hormone (hGH) gene is negatively regulated by triiodothyronine (T3) treatment in transfected rat pituitary tumor cells. We now demonstrate that this responsiveness mediated a negative thyroid response element (nTRE) localized to the proximal 3'-untranslated/3'-flanking region (3'-UT/FR). This binds receptors specifically and with high affinity. nTRE function was promoter-dependent, since it suppressed activity of positive TRE chorionic somatomammotropin promoter, partially repressed herpes simplex virus TK but did not actin or Rous sarcoma promoters. T3 alter transcript termination sites nor selectively affect stability transcripts containing hGH 3'-UT/FR when transcription blocked actinomycin D treatment. depended on its location 3'-UT/FR; inactive positioned down-stream simian 40 (SV40) 3'-UT/FR, acted as placed upstream promoter. These results novel localization unique properties which suggests expanded mechanisms can expression.
