Molecular Cloning and Characterization of a Surface-Localized Adhesion Protein in Mycoplasma bovis Hubei-1 Strain

摘要: Mycoplasma bovis (M. bovis) is an important pathogen that causes various bovine diseases, such as mastitis in cows and pneumonia calves. The surface proteins are generally thought to play a central role the pathogenesis of this organism. We screened entire genome M. Hubei-1 discovered gene named vpmaX encodes 25 kDa variable lipoprotein A (VpmaX). Sequence analysis revealed VpmaX contains several repetitive units typical bacterial signal sequence. was cloned expressed E. coli obtain recombinant (rVpmaX). Western blot using rabbit antibody against rVpmaX demonstrated membrane protein. Immunostaining visualized via confocal laser scanning microscopy showed able adhere embryonic lung cells (EBL), also confirmed by sandwich ELISA. In summary, surface-localized adhesion protein identified Hubei-1.
