摘要: Abstract A method for preparing suspensions of hepatocytes with long-term viability is described. Its originality lies in the use, after conventional preparation, a Percoll density gradient centrifugation which allows complete and rapid separation cell debris released proteases from intact hepatocytes. series functional reactions characterizing drug metabolism ( P -450 level, ethoxycoumarin hydroxylation, methylumbelliferone conjugation) membrane transport integrity (α-aminoisobutyric acid its stimulation by both insulin glucagon) were conducted, parallel to trypan blue exclusion test, evaluate metabolic activity preparations. This investigation clearly demonstrates that use significantly increases (from 5–6 30–35 h) half-life freshly isolated hepatocyte suspensions.
