摘要: Osthole, a major component isolated from the fruit of Cnidium monnieri (L.) Cusson, has been widely used in traditional Chinese medicine. We developed and validated rapid sensitive LC-MS/MS method for quantification osthole rat plasma. Sample preparation involved simple liquid–liquid extraction by ethyl acetate after addition imperatorin as internal standard (IS). The analyte was separated using C18 column with mobile phase methanol–0.1% formic acid (80:20, v/v) at flow rate 0.4 mL/min. elutes were detected under positive electrospray ionization multiple reaction monitoring mode. 0.5 ng/mL lower limit detection. Good linearity obtained over range 1.0–500.0 ng/mL. intra inter-batch accuracy plasma samples ranged 99.5 to 108.1% variation <8.9%. stability, efficiency matrix effect also acceptable.
