New Perspectives in the Studies on Endocannabinoid and Cannabis: 2-Arachidonoylglycerol as a Possible Novel Mediator of Inflammation

摘要: 2-Arachidonoylglycerol is an endogenous ligand for the cannabinoid receptors. To date, two types of receptors (CB1 and CB2) have been identified. The CB1 receptor assumed to be involved in attenuation synaptic transmission. On other hand, physiological roles CB2 receptor, which abundantly expressed several inflammatory cells immunocompetent cells, not yet fully elucidated. Recently, we investigated detail possible 2-arachidonoylglycerol inflammation. We found that induces activation p42/44 p38 mitogen-activated protein kinases c-Jun N-terminal kinase; actin rearrangement morphological changes; augmented production chemokines HL-60 cells; migration differentiated into macrophage-like human monocytes, natural killer eosinophils. also level mouse ear markedly elevated following treatment with 12-O-tetradecanoylphorbol 13-acetate, acute Notably, inflammation induced by 13-acetate was blocked SR144528, a CB2-receptor antagonist. Similar results were obtained allergic model mice. These strongly suggest plays essential stimulation various reactions vivo.
