Homotypic Interactions Mediated by Slamf1 and Slamf6 Receptors Control NKT Cell Lineage Development

摘要: Commitment to the T and NKT cell lineages is determined during αβ receptor (TCR)-mediated interactions of common precursors with ligand-expressing cells in thymus. Whereas mainstream thymocyte recognize Major Histocompatibility Complex (MHC) ligands expressed by stromal cells, interact CD1d cortical thymocytes. Here, we demonstrate that such homotypic T-T generate ‘second signals’ mediated cooperative engagement homophilic receptors Ly108 SLAM downstream recruitment adaptor SAP Src kinase Fyn, which are essential for lineage expansion differentiation lineage. These required TCR therefore only occur when selecting presented thymocytes rather than epithelial do not express or SLAM. Thus, topology ligand recognition determines availability a co-signaling pathway development.
