摘要: Abstract Proprotein convertase 7 (PC7) is a member of the subtilisin-like proprotein family, which involved in endoproteolysis variety precursor proteins. Under steady state conditions, PC7 mainly localized trans-Golgi network, but small fraction found at cell surface. So far, no sorting signals for membrane trafficking have been identified PC7. In this study, we examined internalization from plasma membrane. Our results show that mediated by clathrin-coated vesicles. After inhibition clathrin-mediated endocytosis using hypertonic conditions or molecule inhibitor, Pitstop 2, accumulated Furthermore, was present isolated To determine motif, constructs were generated parts N and C terminus cytoplasmic tail deleted, chimeric proteins constructed consisting luminal transmembrane domains Tac (CD25) domain Antibody uptake experiments as well surface biotinylation demonstrated region between Ala713 Cys726 essential sufficient not network localization.
