Human LBP-32/MGR is a Repressor of the P450scc in Human Choriocarcinoma Cell Line JEG-3

摘要: Steroid hormones regulate a wide range of physiologic functions in humans. The cholesterol side-chain cleavage enzyme P450scc regulates the initial step biosynthesis all steroid hormones. We investigated expression by studying potential regulator P450scc, LBP-32/MGR. Using Northern blot, we found that LBP-32/MGR mRNA was expressed mainly human placenta. radiation hybrid mapping, identified on chromosome 2p25. Recombinant protein bound preferentially to DNA fragment from promoter vitro and exhibited clear nuclear localization transfected cells. Luciferase reporter gene assays showed specifically repressed transcriptional activation promoter. Because placental is essential for pregnancy biosynthesis, repressor activity JEG-3 cells suggest it has role as modulator biosynthesis.
