Alterations in Fatty Acid Utilization and an Impaired Antioxidant Defense Mechanism Are Early Events in Podocyte Injury

摘要: Ultrastructural alterations of podocytes are closely associated with loss glomerular filtration function. In the present study, we explored changes at proteome level that paralleled disturbances podocyte architecture in early stages puromycin aminonucleoside (PA) nephrosis vivo. Using two-dimensional fluorescence difference gel electrophoresis and vacuum matrix-assisted laser desorption/ionization mass spectrometry combined postsource decay fragment ion analysis high- energy collision-induced dissociation tandem spectrometry, 23 differentially expressed protein spots, corresponding to 16 proteins involved various cellular functions, were unambiguously identified, a subset was corroborated by Western blot analysis. The majority these primarily related fatty acid metabolism redox regulation. Key enzymes mitochondrial β-oxidation pathway antioxidant consistently down-regulated PA nephrosis. These increased expression levels CD36. treatment murine culture resembled specific vitro. this cell system, modulatory effects albumin-bound acids on Mn-superoxide dismutase response demonstrated as well. Taken together, results indicate disrupted concert an impaired defense mechanism may play role PA-induced lesions podocytes.
