摘要: During myelin formation, membrane-associated proteins have to be sorted and transported in specified membrane regions such as compact non-compact membranes. One protein that may involved a process is the Myelin Lymphocyte MAL (VIP17/ MVP17). was identified novel component expressed by oligodendrocytes Schwann cells. Since has been shown important apical sorting machinery of polarized cells, we started investigate possible functional role molecules. In this study, generated cDNA constructs with green fluorescent (GFP) either at N- or C-terminus MAL. Transfection experiments showed GFP-MAL expression resembles normal MAL, whereas MAL-GFP fusion construct not properly within cell. Furthermore, could demonstrate enriched detergent insoluble glycolipid-enriched microdomains already seen for untagged As prerequisite generation transgenic mice expressing under control its own regulatory elements, an 8-kb promotor fragment fused GFP-MAL. Oli-neu oligodendrocyte cell line expressed, but only which were stimulated differentiation cAMP. summary, results confirm incorporated into detergent-insoluble complexes sufficient activated oligodendrocytes.
