Comparison of enzyme-linked immunosorbent assay and indirect fluorescent antibody test for the detection of IgG antibodies to sarcocystis muris

摘要: Abstract Sarcocystis muris cystozoites were separated from host tissue after mechanical mincing and homogenization by discontinuous density gradient centrifugation using Percoll®. The isolated antigen, consisting of more than 95% cystozoites, was used in an enzyme-linked immunosorbent assay (ELISA) indirect fluorescent antibody test (IFAT). In the ELISA, antigen at a very low protein concentration 0.44 μg/ml. Good reproducibility results achieved with different conjugates reference sera when measured optical densities converted into index related to standard. From examination 500 non-infected mice, 0.300 serum dilution 1:10 determined as threshold for positive reaction ELISA. Specific antibodies first detected between 18 49 days experimental infection (dpi) S. muris. ELISA well IFAT highly sensitive about 50 dpi onwards up end period (182 dpi). However, unfixed appeared before specific 11 25 starting fluorescence apical pole cystozoite resulting bright whole 32 or 35 onwards. Both serotests showed only slight crossreactions high titred Toxoplasma gondii 1:10. activity lasted least 13 months eight IFAT.
