摘要: Lec13 cells, a variant Chinese hamster ovary cell line, were used to produce human IgG1 that deficient in fucose attached the Asn297-linked carbohydrate but otherwise similar found produced normal lines and from serum. Lack of on had no effect binding FcγRI, C1q, or neonatal Fc receptor. Although change affinity was for His131 polymorphic form FcγRIIA, slight improvement evident FcγRIIB Arg131 FcγRIIA form. In contrast, fucose-deficient FcγRIIIA improved up 50-fold. Antibody-dependent cellular cytotoxicity assays using purified peripheral blood monocytes natural killer cells several donors showed enhanced cytotoxicity, especially at lower antibody concentrations. When combined with an protein exhibited antibody-dependent lack synergistic.
