Two Novel Curcumin Analogues Induced Reactive Oxygen Species Generation and Mitochondrial-Related Apoptosis in Human Breast Cancer MCF - 7 Cells

摘要: Objective: Curcumin has been shown to have significant protective effects against cancer and induction of apoptosis is a crucial strategy for therapy, so we now evaluated the mechanisms involved in two novel asymmetric curcumin analogues induced cell death MCF - 7 cells. Methods: The cytotoxicity towards tumor cells was investigated by MTT assays. morphological analysis using laser scanning confocal microscope. Futher cycle analysis, reactive oxygen species (ROS), mitochondrial transmembrane potentials (Δφm), intracellular Ca2+ levels assays via flow cytometry (FCM). We used western blot determine expressions apoptosis-related factors p38MAPK at protein level. Results: showed loss viability, reduced membrane potential increased levels, production ROS, which activated pro-apoptotic p38 mitogen-activated kinase. Pretreatment with antioxidant, N-acetylcysteine, inhibited both mediated ROS cytotoxicity. Western blotting revealed that Δφm Bcl-2, Bax Bak expression; this promoted release cytochrome c inducing factor from mitochondria cytosol, activation caspase-9 caspase-3 apoptosis. Conclusion: displays strong antitumor effect through ROS-dependent pathway cells, promising be developed as compounds.
