摘要: Genes SNO1 and SNZ1 are Saccharomyces cerevisiae homologues of PDX2 PDX1 which participate in pyridoxine synthesis the fungus Cercospora nicotianae. In order to clarify their function, two genes were expressed Escherichia coli either individually or simultaneously with without a His-tag. When simultaneously, protein products formed complex showed glutaminase activity. purified homogeneity, exhibited specific activity 480 nmol.mg(-1).min(-1) as glutaminase, Km 3.4 mm for glutamine. These values comparable those other glutamine amidotransferases. addition, was impaired by 6-diazo-5-oxo-L-norleucine time- dose-dependent manner enzyme protected from deactivation data suggest strongly that Sno1p Snz1p is amidotransferase former serving although barely detectable alone. The function amido acceptor ammonia remain be identified.
