摘要: Human growth hormone (hGH) is frequently used clinically for abnormalities in children and also adults with deficiency. The usually administered to the individuals by frequent injections. In present study we investigated potential of bone marrow stromal cells as vehicles deliver GH vivo infusion transduced hGH cDNA into mice femurs. effect on vitro was assessed. Bone established from a mouse model human osteogenesis imperfecta (oim) were retrovirus containing neomycin resistance genes. hGH-expressing selected medium G418 then assessed expression vitro. synthesized 15 ng/10(6) per 24 h exhibited alkaline phosphatase activity when they treated recombinant morphogenetic protein 2 (rhBMP-2). proliferated faster than LacZ but did not exhibit higher rate matrix synthesis. When x 10(6) hGH+ injected femurs mice, detected serum recipient up 10 days after injection. highest level expression, 750 pg/ml, I day injection cells. conditioned that flushed at 1, 3, 6 cell These data indicate could potentially be therapeutically delivery or any other therapeutic proteins targeted bone. suggest may exert its effects increasing their proliferation.
