A role for Dimethylarginine Dimethylaminohydrolase 1 (DDAH1) in mammalian development

摘要: Nitric oxide has been linked to a number of embryonic processes, yet the role nitric signalling in development remains largely unknown. Dimethylarginine Dimethylaminohydrolase 1 and 2 (DDAH1/2) catalyse breakdown asymmetric dimethylarginine, an endogenous inhibitor production, may also have oxide-independent functions. We generated transgenic mice targeting DDAH1 DDAH2 loci. Here we report that homozygous null embryos are at low frequency, do not progress through development. During normal RNA is expressed left ventricle, cardiac outflow tract developing vasculature. In contrast, viable fertile, with lifespan. expression seen ventricle tract, additionally peripheral nervous system. Both limb buds patterns overlapping areas high synthase activity. These implicate development, possibly specific effects on pathways.
