Treatment of licorice seeds with colchicine: changes in seedling DNA levels and anthocyanin and glycyrrhizic acid contents of derived callus cultures.

摘要: The use of colchicine to induce polyploids increases secondary metabolite production potential and has been used for many years the valuable compounds in plants. This project took advantage this method increase metabolites licorice. For purpose, seeds licorice, Glycyrrhiza glabra var. glandulifera, were treated with different concentrations 24 hours then cultivated vitro. After a month, effect on cellular DNA level cotyledons was analyzed by spectrophotometry flow cytometry. callus induction, root explants one month old plantlets derived from transferred MS medium containing growth regulators anthocyanin glycyrrhizic acid levels tissues measured after two months growth. total content 0.05%, 0.08% 0.1% increased significantly. Treated plants had numbers larger stomata, significantly those hours. treatment, root, shoot leaf thickness found be increased, while their length decreased. Results cytometry showed changes ploidy obtained treatment (mixoploids) (tetraploids) colchicine. Anthocyanin amount all treatments especially 0.1 0.03% seems prove an polyploid licorice tissues.
