摘要: The notion of using synthetic heterocycles instead the native bases to interface with DNA and RNA has been explored for nearly 60 years. Unnatural compatible DNA/RNA coding have potential expand genetic code co-opt machinery biology access new macromolecular function; accordingly, this body research is core biology. While much literature on artificial focuses expansion, there a significant growing effort docking noncoding nucleic acid interfaces; approach seeks illuminate major processes acids, including regulation transcription, translation, transport, transcript lifetimes. These avenues at interfaces in common fundamental principles molecular recognition. Herein, we provide an overview foundational biophysics base recognition unnatural context developing area targeting bases, focus systems developed through iterative design biophysical study.
