摘要: The mouse gene encoding adrenal steroid 11β-hydroxylase (11β-OHase) has been cloned and the nucleotide sequence of its 5′ end determined. coding regions sequenced are homologous (75%) to bovine 11β-OHase cDNA. 5′-flanking region contains a potential cAMP response element (TGACGTGA) located 56 base pairs upstream transcription initiation site (position −56) two motifs at positions −249 −148 which similar an postulated be required for expression 21-hydroxylase. Transfection Y1 adrenocortical tumor cells MA-10 testicular Leydig with plasmids containing promoter linked growth hormone reporter showed that can direct cell-specific expression. Deletion analyses suggest multiple elements, one is between −425 −338 second −123, interact produce full levels activity. Mutant defective in cAMP-dependent protein kinase activity do not express driven by promoter, indicating requires intact messenger system. Moreover, mutation putative position −56 abolishes These experiments thus present useful system investigation cis-acting elements involved transcriptional regulation 11β-OHase.
