Multiple regulatory elements determine adrenocortical expression of steroid 21-hydroxylase.

摘要: Steroid 21-hydroxylase (21-OHase) is specifically expressed at high levels in the adrenal cortex, where it required for synthesis of mineralocorticoids and glucocorticoids. In this study, we have investigated regulatory elements 21-OHase promoter region which contribute to expression gene Y1 adrenocortical cells. Eight potential 5'-flanking were identified by DNase I footprinting gel mobility shift experiments. Some these footprints produced nuclear extracts from many cell lines, whereas other interactions seen only when using MA-10 Leydig tumor Mutation most markedly decreased a transfected into cells, thus documenting their functional importance expression. Moreover, oligonucleotides containing sequences two related -65 -210, share heptamer AGGTCAG, increased activity heterologous cell-specific manner. Collectively, results demonstrate that cells requires among multiple cis-acting proteins.
