摘要: We report a sensitive, generic method for quantitative profiling of bile acids and other endogenous metabolites in small quantities various biological fluids tissues. The is based on straightforward sample preparation, separation by reversed-phase high performance liquid-chromatography mass spectrometry (HPLC-MS) electrospray ionisation the negative mode (ESI-). Detection performed full scan using linear ion trap Fourier transform spectrometer (LTQ-FTMS) generating data many (endogenous) metabolites, not only acids. A validation urine, plasma liver was 17 including their taurine, sulfate glycine conjugates. 0.01-1 μM range. accuracy human ranges from 74 to 113%, urine 77 104% mouse 79 140%. precision 2 20% pooled samples even studies with large number (n > 250). successfully applied multi-compartmental APOE*3-Leiden study, main goal which analyze effect increasing dietary cholesterol concentrations hepatic homeostasis acid synthesis. Serum different treatment groups were profiled new method. Statistically significant differences between diet observed regarding total as well individual concentrations. © 2008 Elsevier B.V.
