摘要: We have determined the complete nucleotide sequence of gene for crown gall enzyme, octopine synthase. The was derived from cloned fragments Agrobacterium tumefaciens Ti plasmid Ach5. It displayed a continuous open reading frame encoding polypeptide chain 358 amino acids. positions corresponding to 5' end and poly(A) addition site mature synthase mRNA tobacco tumor cell line were by S1 nuclease mapping. Two sequences closely resembling transcriptional control regions found in eukaryotic genes transcribed RNA polymerase II identified flanking genomic DNA: 5'-TATTTAAA-3' located 32 base pairs upstream initiation transcription, hexanucleotide 5'-AATAAT-3' occurred 17 front site. No Shine-Dalgarno present untranslated leader sequence. observations indicate that this DNA sequence, although naturally carried bacterial plasmid, is programmed as functional plant gene.
