Promoter methylation of INK4a/ARF as detected in bile-significance for the differential diagnosis in biliary disease.

摘要: Purpose: There is a need to enhance endobiliary cytotechniques by molecular marker lesions. This of special significance for patients with primary sclerosing cholangitis, disease predisposing the development cholangiocarcinoma. The INK4a/ADP ribosylation factor (ARF) locus encodes two tumor suppressor genes: p16INK4a and p14ARF. has been shown be major in Experimental Design: In an effort evaluate potential diagnostic role p14ARF promoter methylation biliary disease, endoscopical obtained bile specimens 71 were analyzed (26 choledocholithiasis, 6 normal results, 23 duct carcinoma, 5 gall bladder carcinoma). Eleven cholangitis enrolled. Results: Merely 6% (2 32) from without evidence malignant but 53.5% malignancies (15 28) showed p16 (p14: 3 46.2%, respectively). concordance rates detected either or tissue was high. similar prevalence as disease. Conclusions: study demonstrates: (a) high frequency specificity INK4a/ARF compared mere cholangitis; (b) capability detect these alterations reliably endoscopically bile. Thus, INK4a/ARF’s status represents candidate endoscopic diagnosis
