Succinate decarboxylation was measured by the formation of 14CO2 1,4-14C-succinate particle free, dialysed mitochondrial extract liver fluke. It has an absolute requirement Mg2+ CoA. ATP, ADP inorganic phosphate are essential optimal activity. Ap5A, inhibitor adenylate kinase, glutathione also necessary. GTP supports as well provided is present. The CO2 propionate greatly exceeds amount CoA initially present reaction mixture. A net, substrate-level phosphorylation occurs, formed being equivalent to production or propionate. This inhibited flukes incubated vitro with mebendazole. concluded that required spark fermentation when initial substrate intermediate substrates absent; terminal step catalysed transferase which transfers propionyl succinate; coupled methylmalonyl-CoA. scheme presented.
