Dynamic Development of Fecal Microbiome During the Progression of Diabetes Mellitus in Zucker Diabetic Fatty Rats.

摘要: Background: Although substantial efforts have been made to link the gut microbiota type 2 diabetes, dynamic changes in fecal microbiome under pathological conditions of diabetes not investigated. Methods: Four male Zucker diabetic fatty (ZDF) rats received Purina 5008 chow [protein = 23.6%, Nitrogen-Free Extract (by difference) 50.3%, fiber (crude) 3.3%, ash 6.1%, fat (ether extract) 6.7%, and (acid hydrolysis) 8.1%] for 8 weeks. A total 32 stool samples were collected from weeks 15 four rats. To decipher microbial populations these samples, we used a 16S rRNA gene sequencing approach. Results: Microbiome analysis showed that associated with age disease progression. In all stages weeks, phyla Firmicutes, Bacteroidetes, Actinobacteria, Proteobacteria primarily dominated Lactobacillus Turicibacter predominant genera 8- 10-week-old rats, Bifidobacterium, Lactobacillus, Ruminococcus, Allobaculum most abundant 15-week-old Of interest, compared earlier relatively greater diversity (at genus level) was observed at 10 age. 12-week-old had highest diversity, 13-15-week-old reduced. Spearman's correlation F/B negatively correlated Random blood glucose but positively Ruminococcus Simpson's index. Conclusion: We demonstrated time-dependent alterations abundance during progression ZDF At level, observed. believe this work will enhance our understanding development help further analyze role metabolic diseases. Furthermore, may also provide an effective strategy clinical treatment through intervention.
