摘要: The present investigation evaluates the cardiovascular effects of anorexigenic mediator alpha-melanocyte stimulating hormone (MSH), in a rat model type 2 diabetes. Osmotic mini pumps delivering MSH or vehicle, for 6 weeks, were surgically implanted Zucker Diabetic Fatty (ZDF) rats. Serum parameters, blood pressure, and weight gain monitored along with oral glucose tolerance (OGTT). Echocardiography was conducted and, following sacrifice, treatment on ischemia/reperfusion cardiac injury assessed using isolated working heart method. Nicotinamide adenine dinucleotide phosphate (NADPH) oxidase activity measured to evaluate levels oxidative stress, force measurements performed cardiomyocytes determine calcium sensitivity, active tension myofilament co-operation. Vascular status also evaluated arterioles contractile measurement setup. echocardiographic parameters ejection fraction (EF), fractional shortening (FS), isovolumetric relaxation time (IVRT), mitral annular plane systolic excursion (MAPSE), Tei-index significantly better MSH-treated group compared ZDF controls. Isolated aortic coronary flow increased treated rats, higher Hill coefficient indicated co-operation group. We conclude that improves global functions but these are not related vascular status.
