Renal Protective Effects of Low Molecular Weight of Inonotus obliquus Polysaccharide (LIOP) on HFD/STZ-Induced Nephropathy in Mice

摘要: Diabetic nephropathy (DN) is the leading cause of end-stage renal disease in diabetes mellitus. Oxidative stress, insulin resistance and pro-inflammatory cytokines have been shown to play an important role pathogeneses damage on type 2 mellitus (DM). Inonotus obliquus (IO) a white rot fungus that belongs family Hymenochaetaceae; it has used as edible mushroom exhibits many biological activities including anti-tumor, anti-oxidant, anti-inflammatory anti-hyperglycemic properties. Especially water-soluble polysaccharides (IOPs) previously reported significantly inhibit LPS-induced inflammatory mice protect from streptozotocin (STZ)-induced diabetic rats. In order identify nephroprotective effects low molecular weight IOP fraction (LIOP), fruiting bodies obliquus, high-fat diet (HFD) plus STZ-induced 2-like C57BL/6 were investigated this study. Our data showed eight weeks administration 10–100 kDa, LIOP (300 mg/kg) had progressively increased their sensitivity glucose (less tolerance), reduced triglyceride levels, elevated HDL/LDL ratio decreased urinary albumin/creatinine ratio(ACR) compared control group. By pathological immunohistochemical examinations, was indicated can restore integrity glomerular capsules increase numbers mesangial cells, associated with expression TGF-β cortex mice. Consistently, three days (100 μg/mL) incubation also provided protection against STZ + AGEs-induced glucotoxicity tubular cells (LLC-PK1), while levels NF-κB dose-dependent manner. findings demonstrate treatment could ameliorate glucolipotoxicity-induced fibrosis, possibly partly via inhibition NF-κB/TGF-β1 signaling pathway
