Cloning of Notl-linked DNA Detected by Restriction Landmark Genomic Scanning of Human Genome

摘要: Epigenetic alterations are common features of human solid tumors, though global DNA methylation has been difficult to assess. Restriction Landmark Genomic Scanning (RLGS) is one technology examine epigenetic at several thousand Notl sites promoter regions in tumor genome. To assess sequence information for sequences RLGS gel, we cloned 1,161 unique Notl-linked clones, compromising about 60% the spots soluble region profile, and performed BLAT searches on UCSC genome server, May 2004 Freeze. 1,023 (88%) were matched CpG islands showing a large bias toward identifying potential genes or islands. The Notl-loci had high frequency (71%) occurrence within near 5' ends known rather than 3' intragenic regions, making potent tool identification gene-associated events. By mixing gels with all addressed 151 onto standard gel compared them previous reports from types tumors. We hope our will be useful identify novel targets any
