The growth and metastasis of human, HER-2/neu-overexpressing tumor cell lines in male SCID mice.

摘要: HER-2/neu is overexpressed on a variety of human adenocarcinomas and overexpression has been associated with poor prognosis. For this reason, HER-2 become an attractive target for immunotherapy. To facilitate testing anti-HER-2-monoclonal antibodies (MAbs) immunotoxins (ITs), we have evaluated the in vivo growth metastatic spread three HER-2-overexpressing breast cancer cell lines (BT474, MDA-MB-453 HCC1954) one ovarian line (SKOV3.ip1) pre-irradiated male SCID mice using subcutaneous (s.c.), intravenous (i.v.) intraperitoneal (i.p.) routes injection. All tested grew as s.c. tumors BT474 cells after injection was improved by co-inoculation Matrigel. Metastases to lungs were detectable PCR or histopathology much lesser extent HCC1954, MD-MB-453 SKOV3.ip1cells. I.P. HCC1954 SKOV3.ip1 produced fatal ascites while i.v. SKOV3.ip1, but not cells, resulted infiltration death within 9–11 weeks.
