摘要: Receptors for thiazide diuretic drugs in the rat renal cortex have recently been identified through binding of [3H]metolazone, a potent with thiazide-like mechanism action. The present studies describe rapid and reversible alterations that occur receptors following acute ischemia rat. apparent density kidney membranes as measured by [3H]metolazone was reduced 90% 10 min produced clamping pedicle. With release clamp subsequent reperfusion min, receptor returned to within 40% control levels. Ischemia did not alter affinity [3H]-metolazone. Sections prepared from incubated oxygenated media vitro displayed similar changes receptors. Hypoxia 10- 30-min duration incubating sections nitrogen-saturated caused significant decrease return media. Similar decreases were obtained mitochondrial inhibitors, dinitrophenol rotenone, but ouabain. These results indicate undergo form regulation controlling mechanisms are dependent on metabolic energy.
