摘要: Idiopathic myelofibrosis (IM) is characterized by the constitutive mobilization of CD34+ cells. IM peripheral blood (PB) cells had a reduced cloning efficiency and lower frequency cobblestone areas compared with normal granulocyte colony-stimulating factor (G-CSF)-mobilized PB engrafted nonobese diabetic/severe combined immunodeficient (NOD/SCID) mice, demonstrating that they contain bone marrow (BM)-repopulating G-CSF-mobilized produced multiple hematopoietic lineages within NOD/SCID mice predominance CD19+ By contrast, predominantly CD33+ cells, increased numbers CD41+ but fewer Transcriptional clonality assays human demonstrated their clonal origin. from one patient isolated prior to leukemic transformation were capable generating acute leukemia in mice. The exhibited same abnormal karyotype as primary portion population. These findings demonstrate BM-repopulating more differentiated progenitor are constitutively mobilized into IM, differentiation program abnormal. In addition, model may be useful gaining an understanding events occurring during transition leukemia. (Blood. 2005;105:1699-1705)
